Atomic Emission Spectrophotometer (Perkin Elmer 5100 AA)

Turn on air and acetylene gas.
Turn on computer. Turn on AA.
Double click the icon which says AA win lab analyst.
Press ignore twice.
Choose the technique: Flame
To open existing methods go to File - Open -Method. Then select the desired method from the menu.
To create new methods go to File - New -Method. A box will appear saying Create New Method on top. Select the following:
Select the element to be determined from the menu.
Start with : Recommended values
Give the new method a name
Press ok
You will see a box saying method editor on top. Select the following:
Identification: Write method description
Spectro method: The value of wavelength and slit width are selected by default. Modified Setting:No
Signal: Select the desired type: Atomic Absorption
Emission
Background Correction AA
Background Absorption
Timing: Read Time(Sec):5.00
Real Time (sec): 0.00
Flame Type: Select Air/C2H2
Oxide Flow (L/min): 10:00
Fuel Flow (L/min):2:00
Modified Setting: No
Then go to calibration:
Select the appropriate calibration equation
Max decimal places:3, Max significant figure: 4
Units: calibration: mg/L, sample: mg/L
Replicates: Number-fixed 3
Next go to Standard Concentration:
Put IDs and concentrations for blanks and standards in the table.
For emission put the concentrations from low to high.

To save the new files go to File - Save -Method.
Click on Flame icon. Click on the switch to turn on the flame. Click it twice if needed.
When the flame is stable put the hose in deionized water.
Lamp is not required in emission method.
Open three windows: Result, peak and calibration.
Open the manual sampling window.
ASPIRATION- The pop up menu will give you instruction to aspirate the standard with highest concentration first. As emission method does not require a lamp it sets up the wavelength from the highest concentration standard.
The pop up menu will give you instruction to aspirate the blank. You have to wipe off the hose and run the blank in between analyzing each standard and sample.
Then analyze standards from lowest to highest concentration. You will see the calibration curve in the calibration window.
After running the standards you can run the samples. You have to watch that your sample concentrations fall in range between the lowest and highest concentration of your calibration curve. If it goes above the highest concentration of your standard you may have to dilute your samples.
When your done turn off flame and bleed gas.
Turn off computer and AA.
Turn of gas (air & acetylene).
