
Turn on the monitor, computer and spectrophotometer. The instrument has to be warmed from half an hour to one hour before it can be ready for use. Select Windows Start>Programs>Shimadzu>UVProbe or double-click 0n UVProbe icon on the desktop.
Connect to the spectrophotometer: Select a module. Select Window>Spectrum, Kinetics or Photometric before the spectrophotometer can be connected.
Ensure the spectrophotometer is on and then click the connect button. When connection with the instrument is established the Connect button changes to Start and photometer buttons become active. You can begin taking records.
Perform a baseline correction:
Click Baseline on Photometer button bar to initiate the baseline operation. Enter 700 as the start and 300 as the End parameters. Then click OK.
Create a Data Collection Method:
Select Edit>Method or click the Method icon to display the Spectrum Method dialogue box.
Set the Wavelength Range; enter 600 in the Start box and 450 in the End box.
Scan Speed: Select Medium.
Sampling Interval List: Select 1.
Scan Mode: Select Single.
Select Instrument Parameters tab. Measuring Mode: Select Absorbance. Then click OK.
Save a Data Collection Method:
Select File>Save as.
Save the method as .smd file.
Collect the Data:
Place the sample and reference into the sample and reference compartments of the spectrophotometer.
Click the Start button on the Instrument bar to initiate the Scan.
When the scan is complete enter the file and data storage name in the New Data Set dialog box.
Save the Data:
Select File>Save as.
Save data as .spc file.
